熒光抗體染色標本的固定在熒光抗體試驗中，染色標本的固定是個很重要的步聚。通過固定，不僅要使標記的抗體易于接近抗原，從而發生反應，而且要求標本中的抗原的活性不受損失，同時還要保護其自然形態和位置。因此，根據所研究的抗原和組織細胞種類的不同，相應地采用不同的固定方法。應用zui廣泛的固定液是丙酮和乙醇，其次是甲醇、甲醛、丙烯醛等。切片標本大多用乙醇固定，組織培養標本主要用丙酮固定。固定液的濃度：丙酮為100%，乙醇為100%或95%，甲醛為10%，甲醇為100% 。固定的溫度和時間變化很大，溫度從-70℃至37℃都有應用，時間一般在10-30min。
